The stand-in skipper was caught at slip on 128 with two overs until the close of play, as Danish Kaneria claimed two wickets in the over to finish with 3-73.
Earlier in the day, Mouhammad Yousuf toiled hard for 203 as Pakistan piled on a gritty 445 in response to the England's first innings total of 528-9 declared.
Yousuf reached his double century, only the second Pakistan batsman to do so in a Test at Lord's, before succumbing to England quick Steve Harmison after edging through to keeper Geraint Jones.
Harmison was the pick of the bowlers for England on a day dominated once again by the batsman, the paceman finishing with a haul of 4-94.
However, Strauss was the star of the show for the hosts as he notched up his near-flawless century.
The skipper was particularly productive with the cut shot, plus some neat leg glances and became just the third England captain to score a century in his first match at the helm.
